New · RNA–LNP Mixture–Process DOE
Lipid molar fractions sum to 100%. Standard factorial/CCD designs cannot treat these fractions independently. Use our specialized four-lipid constrained mixture worksheet with process factors, QC-response matching and Project Hub integration.
Open Mixture–Process DOE Studio ↗1 · Design your experiment
Enter two to five independent numeric factors with feasible low/high values. Responses can be RNA capping efficiency, LNP encapsulation, cell-expression signal or assay outcomes.
Numeric factors
Units and limits appear in the run sheet. Use independent factors; constrained composition percentages that must sum to 100% require a mixture design instead. Open LNP mixture DOE ↗
2 · DOE planning checklist
- Define experimental unit: an independent formulation batch, RNA synthesis, biological donor or culture—not technical repeat wells alone.
- Set response and criteria first: state measurement units, method, direction of improvement and acceptable range.
- Protect randomization: consider day, instrument, operator, cell passage, and raw-material lots as possible blocks or covariates.
- Control confounding: fractional factorials alias effects; consider fold-over or follow-up confirmation before mechanistic conclusions.
- Check curvature: factorials with center points can screen curvature, but a quadratic model requires a suitable response-surface design.
- Validate predictions: replicate the proposed optimum and compare to a predefined control before making claims.
DOE in the connected NEXUS pipeline
Project saves include the DOE plan, entered responses, numerical model summary and key caveats, but do not automatically ingest outcomes from the other modules.
3 · Randomized experimental worksheet
Generate a design, then record one primary numerical response per experimental run. Run IDs remain stable during CSV export/import; the experimental order is randomized but reproducible from the seed.
No design generated yet.
4 · Response-model interpretation
Uses coded factors. Full factorial designs fit main and pairwise interactions; half fractions fit main effects only; response-surface designs also include quadratic terms.
Enter responses in the run sheet, then analyze.
5 · Publicly accessible software & reproducibility
Export the worksheet CSV for independent analysis using established open-source software. This offline NEXUS calculator is independently implemented; it does not run or bundle these external packages.
No licensing claims are made for linked software beyond their public availability; confirm the packages' licenses and versions before embedding any upstream code or using in regulated/commercial workflows.
